Abstract:

Many modern tomato (Solanum lycopersicum) varieties have been selected for uniform ripening (u) light green fruit phenotypes in order to facilitate harvests of evenly ripened fruit. By determining that the U locus on chromosome 10 encodes a Golden 2-like (GLK) transcription factor, SlGLK2, it is clear that this transcriptional regulator determines the extent and distribution of chloroplast development in response to light in developing fruit.  As in other plants, two redundant GLKs, SlGLK1 and SlGLK2, are expressed in tomato and pepper leaves and regulate the development of thylakoid membranes and the synthesis of chlorophyll in response to light.  However, only SlGLK2 is expressed in fruit, suggesting that SlGLK2 has specialized functions that modulate chloroplast development for fruit metabolism.  Over-expressing GLKs increased chloroplast development, photosynthesis gene expression and chlorophyll accumulation in fruit before ripening and elevated carbohydrate and carotenoid contents of the ripened fruit.  In developing fruit, SlGLK2 influences the capacity for chloroplast-associated metabolism, which, thereby contributes to the flavor and nutritional properties of mature fruit. The results suggest that selection of u inadvertently compromised ripe fruit quality in exchange for desirable production traits.
